J'ai eu le malheur de faire un update de DSM sans lire les forums avant et je suis dans la même situation que vous :-(
J'ai contacté le support et voici la réponse qu'ils m'ont donnée. Je doute que ça vous serve car j'ai l'impression que vous vous y connaissez pas mal en informatique, ce qui n'est pas mon cas.
Cela étant, cela pourra peut-être aider les autres.
De mon côté, j'en suis à une nouvelle indexation car cela avait disparu lors de l'update. On verra ce que ça donne.
Voici la réponse du support :
Please refer the steps below to check your settings in Media Server.
1. Make sure the firewall on the DiskStation is not blocking any ports. Do this by going to DSM > Control panel > firewall > ensure it says “if no rules are matched: allow access” at the bottom of the page, and save the firewall rule.
2. Please click the "re-index" button to re-index the media files on WebUI > Control Panel > Media Server > Index Folders.
3. Please uncheck all setting under DS UI > Control Panel > Media server > "Browsing Settings".
4. Please disable MIME under DS UI > Control Panel > Media server > "DMA compatibility".
5. Please make sure disable "restrict access to media server for newly detected UPnP/DLNA devices" under media server > DMA compatibility.
6. Please kindly setup your media device profile to be "your media device" on WebUI > Control Panel > Media Server > > "DMA Compatibility" > Device list.
7. let your media device and DiskStation with the default setting of 1500MTU, afterward connect DiskStation directly to a your media device using an Ethernet cable without going through a router.
8.Do cross test to use Window Media Player and also install XBMC on PC to act as DMA device to stream with DS media server and if same issue?
Also if you use Window PC then download Twonkey media Server to act as DS and stream with your DMA client and see if same issue?
9.Please go to DS UI> Media Server > General > SSDP advertisement interval set to less time, then if it helps?
Please check if the [Firewall] or [Auto Block] in Disk Station have blocked the access of your device. To release the rules of [Firewall], please use a ballpoint pen or straightened paperclip to press and hold the RESET button on the back of the server. After about 4 seconds the server should beep once. Immediately release it when beep is heard.
The RESET button on the Synology Product has following functions:
· To restore "admin" account's password to default value(blank).
· To reset IP, DNS, gateway, and other net interfaces to DHCP
· To reset the UI management port to 5000/5001
· To disable PPPoE
· To disable AutoBlock
· To disable link aggregation
· To disable firewall rules

EDIT : Indexation finie. Et tout est rentré dans l'ordre. Dans mon cas, l'updgrade à DSM 5 avait supprimé les données de l'onglet "Dossier Indexé". J'ai donc remis les infos et fais une ré-indexation. Et cela a fonctionné. Bon courage pour les autres.
